EGCG, a major green tea catechin, inhibits PMA-induced 92-kDa Type ¥³ collagenase activation in OVCAR-3 cells

Abstract
Catechins are key components of teas that have anti-invasive properties. We investigated the effects of (-)-epigallocatechin gallate (EGCG), the most abundant catechin in green tea extract, on phorbol 12-myristate 13-acetate (PMA)-induced 92-kDa Type ¥³ collagenase(MMP-9) activation in OVCAR-3 cells. EGCG inhibits PMA-induced MMP-9 activation in a dose-dependent manner as determined by gelatinolytic zymograpy. c-JUN activation by PMA, as determined with inimunoblotting using phospho-specific c-JUN antibody, also abrogated by EGCG. To assess whether EGCG prevent AP-1 activation, in turn, may down-regulate MMP-9, or AP-1 DNA binding activity was examined by EMSA. EGCG was found to decrease AP-1 binding activity in nuclear extracts of OVCAR-3 cells. EGCG also inhibited the increase of MMP-9 promoter activity induced by PMA. However (-)-epicatechin (EC, negative control) did not significantly affect MMP-9, c-JUN and AP-1 activation. Above results suggested that EGCG may exert at least part of its anti-invasive effect by inhibiting MMP-9 through blocking the activation of c-JUN.
